چکیده

RESULTS. The mature T 8 is a 65-amino-acid peptide encoded by the G 8 gene with an acetylated and a farnesylated–methylated Nand C-terminus, respectively. Purified T 3 8 is similar to T 1 1 in that (1) both are heterogeneous, containing methylated and demethylated T subunits; (2) each demethylated dimer migrates faster than its methylated counterpart during native gel electrophoresis, and the methylation-associated mobility differential is masked by pdc binding; and (3) both dimers bind pdc with the same affinity, and the affinity is reduced threefold by PKA phosphorylation of pdc and twofold by demethylation at the C-terminus of T . T 3 8 differs from T 1 1 in exhibiting lower intrinsic electrophoretic mobility, and the difference is unaffected by either pdc binding or the status of T methylation.

Embryonic markers of cone differentiation

PURPOSE Photoreceptor cells are born in two distinct phases of vertebrate retinogenesis. In the mouse retina, cones are born primarily during embryogenesis, while rod formation occurs later in embryogenesis and early postnatal ages. Despite this dichotomy in photoreceptor birthdates, the visual pigments and phototransduction machinery are not reactive to visual stimulus in either type of photor...

Expression of cone-photoreceptor-specific antigens in a cell line derived from retinal tumors in transgenic mice.

PURPOSE To examine an immortalized mouse retinal cell line (661W) for markers characteristic of photoreceptor cells and validate its photoreceptor origin. METHODS The 661W cells were cloned from retinal tumors of a transgenic mouse line that expresses the simian virus (SV) 40 T antigen under control of the human interphotoreceptor retinol-binding protein (IRBP) promoter. Morphologic, immunocy...

Phosducin Regulates the Expression of Transducin βγ Subunits in Rod Photoreceptors and Does Not Contribute to Phototransduction Adaptation

For over a decade, phosducin's interaction with the betagamma subunits of the G protein, transducin, has been thought to contribute to light adaptation by dynamically controlling the amount of transducin heterotrimer available for activation by photoexcited rhodopsin. In this study we directly tested this hypothesis by characterizing the dark- and light-adapted response properties of phosducin ...

Amino acid and cDNA sequence of bovine phosducin, a soluble phosphoprotein from photoreceptor cells.

Vertebrate photoreceptor cells contain a soluble phosphoprotein, phosducin, which complexes with the beta, gamma subunits of the GTP-binding protein, transducin. Light-induced changes in cyclic nucleotide levels modulate the phosphorylation of phosducin by protein kinase A. The complete amino acid sequence of purified phosducin from bovine retinas was determined by Edman degradation from overla...

Expression of soluble phototransduction-associated proteins in ground squirrel retina.

PURPOSE This study describes the expression and distribution of arrestin, phosducin, and recoverin in the cone-dominant retina of the ground squirrel Spermophilus tridecemlineatus. METHODS mRNA expression was studied by blot hybridization of ground squirrel retinal RNA, with human and murine RNA as controls. The distribution of the gene products in the ground squirrel retina was investigated ...
